Zusammenfassung
Unter Phakomatosen (phakos = die Linse, der Fleck) wird eine Gruppe von Erkrankungen verstanden, die sich an Organen ektodermalen Ursprungs, also z. B. Nervensystem, Auge oder Haut manifestiert. Wir zählen hierzu die Neurofibromatose Typ 1 und Typ 2 sowie die tuberöse Sklerose. Bei dieser kongenitalen Krankheitsgruppe bilden sich u. a. Tumoren. Häufig unter den Phakomatosen subsumiert werden auch Angiomatosen an Geweben neuroektodermalen Ursprungs. Sie haben teilweise ebenfalls Tumoreigenschaften wie die Hippel-Lindau-Krankheit, während andere ausschließlich dysontogenetischen Charakter haben (z. B. Sturge-Weber-Angiomatose). Oft unter den Phakomatosen subsumiert werden auch einige weitere seltene angiomatöse neurokutane Syndrome wie das Klippel-Trenaunay-Weber-Syndrom, die Rendu-Osler-Weber-Krankheit, das epidermale Nävussyndrom und andere. Die nosologische Klassifikation besteht seit knapp 100 Jahren, wird aber möglicherweise durch die jüngsten molekulargenetischen Erkenntnisse neu gefasst werden müssen. Diese Forschungen haben nicht nur faszinierende Einsichten in die Entstehung der Phakomatosen gebracht, sondern lassen darüber hinaus vermuten, dass die mitinvolvierten Gene eine über die Phakomatosen hinausgehende Bedeutung in der Tumorgenese haben.
