Abstract
Two major divisions of approach to the identification of cell constituents may be distinguished: 1) the attempt to show the localisation of substances in situ within the cell, or to estimate some constituent within a single cell; here the techniques of microscopy and microchemistry are of great significance: 2) the attempt to isolate from a tissue a fraction containing only one type of cellular inclusion, e.g. chloroplast; this is usually done on a “large” scale and the more “classical” chemical and biochemical analyses may be applied.
