Control of Maternal Immunity to Trophoblast

Abstract

The cellular mechanisms by which maternal immunity might damage trophoblast are described. The absence of plymorphic Class IHLA protects both cytotrophoblast and syncytiotrophoblast from this damage. The control of NK attack has been studied using the antibodies normally made against the R80K antigen of trophoblast. These antibodies have been shown to block NK activity against target cells, and tropboblast. The monoclonal antibody, BAH, against R80K has been shown to prevent NK activity against human and murine targets, and to be extremely efficient at preventing abortion in three mouse models of abortion/resorption. It is suggested that the combination of the anti-R80K antibody and the lack of Class I prevent maternal damage to the semiallogeneic trophoblast of the conceptus. The presence of the antibody in all successful placentae studied suggests that it is a prime controller of maternal response to the syncytiotrophoblast.
