Pathological effects of cyclosporin A in experimental models

Abstract

Cyclosporin A (CsA) administered either alone or in combination with steroids or azathioprine, is now the immunotherapeutic agent of choice in the management of organ allograft rejection. The prospective value of CsA in the treatment of certain autoimmune diseases is presently being evaluated in numerous clinical trials. However, it has been demonstrated both in animals and man that CsA possesses many adverse side-effects, which include renal and hepatic dysfunction, hypertension, mild tremors, hirsutism, abnormal glucose homeostasis and gingival hypertrophy. Some degree of renal impairment is observed in virtually all patients receiving CsA, and it is this attendant nephrotoxicity which limits its clinical use.
