Cell Disruption and Removal of Insolubles

Abstract

Disruption and separation of insolubles are unit operations used in the initial stages of product recovery/isolation. This paper reviews the main methods available both for cell disruption and removal of insolubles. Main insolubles considered are whole cells, cell debris and particles such as inclusion bodies. Separation and renaturation of inclusion bodies and other recombinant particles pose important challenges to modern process biotechnology. This paper critically assesses some of the main shortcomings of existing methodology used in the large scale today and points out recent novel developments that show potential for scale-up. Emphasis is given to methods that show improved selectivity and to novel process concepts in terms of their suitability for bio-processing.
