Zusammenfassung
Frühere in-vivo-Studien unserer Arbeitsgruppe haben am Beispiel des Cholesterol- gefütterten Kaninchens erstmals gezeigt, daß Pentaerithrityltetranitrat (PETN) vaso- protektive Effekte bei Atherosklerose entfalten kann (Kojda et al. 1995). So bewirkte eine nicht-intermittierende, über 16 Wochen andauernde Behandlung mit PETN nicht nur einen Rückgang der Große intimaler atherosklerotischer Läsionen, sondern konnte auch die Entwicklung einer endothelialen Dysfunktion vollständig aufheben. Der Mechanismus dieser antiatherosklerotischen und gefäßprotektiven Eigenschaften von PETN ist bislang unbekannt. Es wird jedoch vermutet, daß die möglicherweise auch klinisch bedeutsame vasoprotektive Wirkung von PETN unter anderem darauf beruhen könnte, daß dieses organische Nitrat, im Gegensatz zu den anderen organischen Nitraten wie Glyceroltrinitrat oder Isosorbid-5-nitrat, eine nur geringe Tendenz zur Ausbildung einer Nitrattoleranz aufweist. Diese sich im wesentlichen auf klinische Erfahrungen gründende Einschätzung (Dück und Richard, 1990) wurde erst kürzlich durch tierexperimentelle Studien unterstützt (Fink et al. 1996; Kojda et al 1995).
