Abstract

In addition to the attention focused upon it because of its pharmacologie properties, bradykinin has also been studied with regard to its immunochemical characteristics. A small peptide which can be rendered immunogenic—i.e. made to elicit the formation of antibody directed against it—bradykinin has both defined amino acid content and defined amino acid sequence. Equally well-defined bradykinin analogs and fragments have made possible an immunochemical assessment of the antigenically important groups and the conformation of bradykinin as recognized by specific antibody. The first section of this chapter will consider the following: methods by which bradykinin has been rendered immunogenic; immunization of animals with bradykinin immunogens and subsequent assessment of the antibody response to bradykinin; efficacy of methods of quantifying bradykinin by radio-immunoassay; use of the radio-immunoassay in determining the conformation of bradykinin; comparison of important groups of bradykinin recognized by the antibody binding site and the biologic receptor site; and immunologic interrelationships of bradykinin and kininogens.
