Zusammenfassung
Die Synthese von Gallensäuren aus Cholesterin (= Cholesterol) wird von mindestens 17 Enzymen katalysiert, für deren kodierende Gene pathogene Mutationen beschrieben wurden. Die geschätzte Prävalenz der Gallensäurensynthesestörungen liegt bei 1:50.000–1:100.000. Die Defekte werden autosomal-rezessiv vererbt und treten vorwiegend bei Konsanguinität in einer Familie auf. Fast alle manifestieren sich im frühen Kindesalter und lassen sich durch orale Gabe von Gallensäurepräparaten oft bis hin zur Normalisierung von Symptomen, Labordaten und Leberhistologie bessern.
