Abstract

A little over 3 years have elapsed since Yanagisawa and his colleagues48 named and described the chemistry and sustained constrictor activity of the polypeptide (endothelin-1) that accumulates in the supernatant of cultured endothelial cells. During these years, interest in this polypeptide has escalated to such a degree that several hundred papers have now been published on the topic. Other endothelins (endothelin-2, endothelin-3, and mouse β-endothelin) have been identified,49,50 chemically homologous polypeptides have been isolated from the venom of the asp Atractaspis engaddensis,24 and their mode of action has been identified.49,50 In addition, attention has been directed to their possible involvement in a variety of pathological conditions, including asthma, vasospasm, hypertension, renal failure, and myocardial infarction.33,34
