Abstract
It is not known yet if the theme of this conference, Neurotransmitter Regulation, is relevant to the problem of narcotic tolerance and dependence. In 1961, Dr. Dora B. Goldstein and I (1961) proposed a unitary theory of tolerance and dependence, based upon the Jacob-Monod (1961) concepts of repression and derepression of protein synthesis as fundamental mechanisms of biochemical regulation. In the same year, Shuster (1961) advanced virtually the same idea independently. Collier (1965) later advanced a somewhat different but related theory. And subsequently, our ideas were further refined and generalized (Goldstein and Goldstein, 1968).
