Abstract

The complex series of steps leading to the production of adequate numbers of blood cells in animals and humans involve the orderly proliferation and differentiation of the pluripotent and committed hemopoietic progenitors in response to the positive and negative feedback influences of regulatory signals. Both humoral and cell-mediated events play a role in this process. The role of cell-cell interaction in the regulation of hemopoiesis, elegantly demonstrated by the in vivo observations of Trentin, 1 has been firmly established as the result of studies utilizing the in vitro clonal assays for hemopoietic progenitors.2–6 These studies have shown that accessory cell populations (T-cells, monocyte-macrophages, endothelial cells, stromal cells, and so on) that comprise the normal hemopoietic microenvironment, can play a significant role in the regulation many aspects of blood cell production. Although the mechanism(s) by which these accessory cells affect hemopoiesis is not understood, both helper and suppressive roles have been ascribed to these cells. It is generally accepted that the helper influences of these environmental cells are, for the most part, exercised at the earliest stages of progenitor cell growth, preparing the cells for action by “terminal differentiation” hormones and factors.5,7
