FormalPara Synthese – Verteilung – Abbau – EliminationPrimidon wird p.o. appliziert mit einer Bioverfügbarkeit von fast 100 %. Beim hepatischen Abbau entstehen die ebenfalls wirksamen Metabolite Phenobarbital (Barbiturate) und Phenylethylmalonamid. Ca. 40 % werden unverändert im Urin ausgeschieden.

Es wird vielfach empfohlen, anstelle der Primidon- die Phenobarbitalkonzentration zu überwachen.
