Abstract
Tests to screen for estrogenicity and appropriate biomarkers of human exposure are required for epidemiological studies of endocrine disruption. This requirement is addressed here by developing a protocol that investigates bioaccumulated xenoestrogens that are candidates for estrogenicity and assesses their combined estrogenic effect. It comprises two major parts. The first part consists of an exhaustive chemical determination of the compounds under study, including procedures for the extraction, purification, identification, and confirmation of the organochlorine pesticides and the quantification of lipids in the samples. This goal is achieved using a semipreparative high-performance liquid chromatographic (HPLC) separation of xenoestrogens from endogenous hormones and subsequent chromatographic analysis of the fractions. The second part corresponds to the testing of HPLC eluted fractions in the E-Screen test for estrogenicity, which yields the estimation of the total effective xenoestrogen burden of each sample.
