Abstract

Gene therapy involves the introduction of foreign therapeutic genes into humans to treat certain diseases. In current protocols, the expressions of the delivered foreign genes are under the control of a constitutive promoter. However, most genes are regulated under physiological conditions in response to various stimuli, including metabolites, growth factors, and hormones. Constitutive expression of foreign genes may result in cytotoxicity as well as undesired immune responses. In order to further the development of gene therapy, it is essential to regulate the expression of the genes once they are delivered into the body.
