Abstract
Unlike higher organisms, genome of viruses is made up of DNA or RNA, which further may be either single or double stranded. The first step in the isolation of nucleic acid from purified virus preparation involves removal of coat protein of the virus particle followed by precipitation of the nucleic acid by ethanol in the presence of a salt. Yield of nucleic acid is determined using spectrophotometer, and quality can be judged by gel electrophoresis. The nature of virus genome is determined by treating the isolated nucleic acid with various enzymes followed by electrophoresis.
