It has been rumored in the theory of enzyme action that the enzyme forms a combination with the substrate which breaks up to yield the products of the reaction and the free enzyme again. Further assumptions, such as that the velocities of combination of enzyme and substrate and of the decomposition of their compound, follow the law of mass action and permit the deduction of mathematical expressions which connect the velocity of the reaction as a whole, at least in the initial stages, with the concentration of enzyme and substrate.
