Cerebral hypoxia—ischemia in adult mice allows the study of the effects of hypoxia in a partially ischemic territory. This model may be clinically relevant in situations of general respiratory-circulatory failure (e.g., in shock) or ischemic tissue that displays a breakdown in oxygen metabolism during stroke progression. In this chapter, we outline this procedure in detail from ligation of the common carotid to administration of the hypoxic gas. We will also describe outcome measurements using behavioral deficits, Nissl stain infarct quantification, and fluorescein isothiocyanate-dextran (FITC-dextran) perfusion marking. Finally, we will briefly discuss the advantages, limitations, and complications with using cerebral hypoxia-ischemia in adult rodents.
