Abstract

DNA sequencing is the process of determining the order of nucleotides in a DNA fragment. Sequencing of viral genome refers to the determination of order of nucleotides in a viral genome. In order to sequence a viral genome, it needs to be first cloned in a plasmid vector. Sequence data helps to identify, characterize and develop DNA-based diagnostics for the diagnosis of the virus. It also helps to compare with other viral genomes and to establish phylogenetic relationships among distant and closely related virus strains. Chemical degradation and chain termination methods are the initial methods for DNA sequencing. However, at present the chain termination method developed by Sanger is commonly used for sequencing. Besides, next-generation sequencing and next-next generation sequencing methods are also being used at present, especially to diagnose and identify unknown viruses infecting plants.
