The study of Toshchakov and Neuwald analyses Toll-Interleukin-1R Resistance domains (TIRs) at a large scale. The authors use a series of statistical approaches to identify 26414 TIRs in publicly available protein sequence databases, and then hierarchically classify them based on group-specific residue patterns. The functionality of the group-defining pattern residues is reviewed in the light of available biochemical and structural data. One example of identified patterns, shown in the accompanying image, presents the evolutionary conserved, TIRAP/Mal-specific residues involved in the assembly of TLR signaling complexes.
