Abstract
2-(N-acetoxyacetylamino)fluorene (AcO-AAF), a model of ultimate carcinogen of 2-(acetylamino)fluorene (AAF), reacts with DNA to give essentially N-(deoxygua-nosine-8-yl)AAF (for a review, see Grunberger and Weinstein 1979). This substitution induces a conformational change in the vicinity of the adduct, leading to a local denaturation of the double helix (Daune and Fuchs 1976).
