Abstract
Almost all fractionators use some modification of Dr. Cohn’s Method 61 to separate normal serum albumin from plasma. These processes involve selective solubility using a series of precipitation steps with cold ethanol to produce the albumin fraction - Fraction V. In the Cohn process this Fraction V is suspended under specific conditions, filtered and reprecipitated to form the albumin-containing fraction - Fraction V rework (Fig. 1). The teaching in our house had been that the “rework” step was a purification step. This concept paradoxically is true and is not true. There are contaminating salts from components used in the process. On the one hand the rework step reduces the ratio of salts to protein 10 fold (Table 1).
