Summary

This chapter critically examines the evidence for the existence of two classes of cell membrane adenosine receptors in peripheral tissues. This concept has been based on two types of observation. 1) Adenosine receptor activation can have either stimulatory or inhibitory effects on adenylate cyclase activity in isolated cells. 2) A series of adenosine analogues exhibit two different and distinct orders of potency on a range of both biochemical and pharmacological preparations. These observations provide suggestive, but not conclusive, evidence for the existence of two receptor types. The different effects of adenosine on adenylate cyclase activity may represent differentiation of stimulus-effect coupling, rather than two structurally dissimilar receptors. The two distinct orders of potency of adenosine analogues may not be solely due to their different affinities for two types of receptor. They may also be a reflection of the different efficacies of these analogues. Consequently, the development of selective antagonists is necessary in order to establish the existence of two types of adenosine receptor.
