Abstract

Adenosine has a potent depressant action on the firing of central neurons in vivo (Phillis et al., 1979), and in behavioral tests adenosine and its derivatives have been shown to have sedative, hypnotic, anticonvulsant and analgesic activity (Dunwiddie and Worth, 1982; Radulovacki et al., 1982; Yarbrough and McGuffin-Clineschmidt, 1981; Barraco et al., 1983; Barraco et al., 1984; Coffin et al., 1984). An adenosinergic nucleus in the magnocellular area of the ventral hypothalamus with extensively ramifying projections has been proposed on the basis of immuno-histochemical staining for adenosine deaminase (Nagy et al., 1984). The effects of these uptake inhibitors are blocked by the adenosine receptor antagonist caffeine and their action is presumed to be a result of an accumulation of adenosine in the extracellular spaces of central nervous tissues.
