Purines and Hypertension: Interaction with Hydralazine

Abstract

The hypotensive effect of hydralazine (1-hydrazinophthalazine, shown in Fig. 1 with related hydrazines) was first reported by Gross et al. (1950) and shortly afterwards it was shown that the compound was effective in lowering blood pressure in man and could be used in anti-hypertensive therapy (Schroeder, 1951; Page, 1951). The role of hydralazine in the treatment of hypertension increased with the use of beta-adrenergic antagonists, which reduce the major reflex effects of vasodilator-induced hypotension, namely tachycardia and an increase in the activity of the renin-angiotensin system (see Koch-Weser, 1976).
