Serum-free cultivation of lymphoid cells

Abstract

Lymphoid cells offer a unique source of human cell material as samples can be taken in large amounts without harm to the donator. Even in animal studies this might be of crucial importance.

The possibility of growing these cells under serum-free, defined environmental conditions offers significant advantages before cultivation in serum-enriched media. Serum does not only stimulate growth, it even makes the interpretation of results more complicated as a complex, undefined mixture named serum is present.

As has been demonstrated in this article no universally accepted medium for serum-free culture of lymphoid cells has yet appeared. If one such medium should be nominated, it would presumably be the enriched Dulbecco's modified Eagle's medium as modified by Iscove and Melchers). Many media are variations on this theme. Different cell lines have different requirements and the optimal medium for a specific cell line has to be worked out for each case.

In general, transformed cells have simpler requirements than more “normal” variants.

Measuring a range of biochemical parameters and cellular markers it has been demonstrated that no fundamental cellular changes are induced when transferring cells from a serum-enriched medium to a serum-free medium.
