Abstract

Hairy root (HR) cultures have proved to be an interesting platform for the production of biopharmaceutical proteins mainly for their intrinsic genetic stability, fast establishment of transgenic lines and fast biomass accumulation. In this chapter, we will describe the protocols used to obtain HR cultures from different plant species (Nicotiana tabacum, Nicotiana benthamiana and Brassica rapa subsp. rapa) based on Rhizobium rhizogenes infection. These protocols have been successfully used to produce different heterologous proteins ranging from fluorescent reporter proteins to antibodies and vaccine antigens. A particular feature of HR cultures is the possibility of inducing the secretion of the proteins of interest in the culture medium which allows a facilitated recovery and purification of the final product. The most efficient protocols used to enhance recombinant protein secretion will be described with a special focus on a simple two-step procedure for the purification of antibodies from the culture medium.
