Abstract
This chapter presents the technique of tissue sectioning for histopathological examination. The different types of microtomes are used for sectioning of the tissue, and among those the rotary microtome is most commonly used in laboratory. This is a heavy and stable microtome, and good-quality 2–3-μm-thin section is possible by it. The chapter discusses about the various types of knives and blades that are used in the microtomy. Nowadays, disposable blades of low- and high-profile types are used in the microtomy. These blades do not need any sharpening and are easy to replace within seconds. The stepwise technique of tissue microtomy along with the various sectioning artefact is also described in detail.
