Analysis of GAPDH and Nucleic Acid Interaction

Abstract

Experimental evidences indicate that the interaction between GAPDH and nucleic acids is as diverse as its interaction with proteins. The nucleic acid binding activity of GAPDH has been associated with tRNA transport, DNA repair, transcription and binding with viral RNAs, telomeric DNA and so on. This chapter will describe some of the validated methods for the analysis of GAPDH–nucleic acid interactions. In general, the methods involve the use of probes (e.g. radiolabeled nucleotide sequences). A large majority of reports use the electrophoretic mobility shift assay with radiolabelled probes, as this offers very robust validation and avoids the need for additional steps in signal detection. Although, recent advances provide the option of using nonradioactive probes, the selection and choice of probe labeling highly depends upon the user and the nature of the analysis.
