Indications for the Chloroplast as a Tri-Compartment System: Micro-Electrode and P515 Measurements Imply Semi-Localized Chemiosmosis

Abstract

The electric response of a thylakoid membrane to a single turnover saturating light flash differed if measured by the electrochromic absorbance change (P515) from the response as measured with the micro-electrode. The slower kinetic components in the P515 response being absent in the micro-electrode measurements (Vredenberg 1981). These slower kinetic components, called reaction II (Schapendonk et al. 1979), in the P515 response were shown not to be a response to a secondary charge separation, known as the Q-cycle (Van Kooten et al. 1983). Because of the difference between micro-electrode and P515 measurements, it was postulated that reaction II was an intramembraneous electric event (Westerhoff et al. 1983).
