Preparation and properties of isolated hepatocytes from sheep

Abstract

The advent of a reliable method for the preparation in high-yield of viable hepatocytes from the rat, developed by Berry and Friend [1], opened up many new studies that otherwise could only be addressed using liver slices (inferior) or perfused liver (ignoring vascular effects). The situation for metabolic studies in larger animals such as the sheep, was even more a problem as sheep liver perfusions were technically cumbersome. Thus the idea of adapting the method of Berry and Friend [1] to prepare lamb, and then later, sheep liver cells was very attractive. Such a method had the potential to allow investigators an opportunity to study the metabolic changes that occurred in ovine liver as weaning proceeded and metabolism changed from non-ruminant to ruminant.
