Molecular Diagnostics for Loma Salmonae and Nucleospora Salmonis (Microsporidia)

Abstract

Microsporidia are some of the smallest intracellular eukaryotic parasites. Some 14 genera of these ubiquitous spore-forming protists have been reported from fishes (Canning and Lom, 1986), with several causing disease in farmed and wild fish populations (reviewed in Shaw and Kent, 1999). Their tiny size (about 2–5 µm long), cryptic habitat within host cells, and difficulty with their in vitro culture necessitates development of molecular tools for highly sensitive and specific diagnostic tests. The traditional utility of electron microscopy for species identification of microsporidians is being superceded by immunological (Luna et al., 1995; Van Gool et al., 1993) and particularly PCR-based diagnostic tools (Da Silva et al., 1996; Hung et al., 1998; Khattra et al., 2000; Malone and McIvor, 1995; Sparfel et al., 1997; Velásquez et al., 1999; Zhu et al., 1993).
