Genetic Fingerprinting of Coffee Leaf Rust Differentials with RAPD Markers

Abstract

Perennial plantation crops such as coffee have a long breeding and selection cycle. Hence, genetic analysis of such plants by conventional methods is a difficult task. Discerning segregating genes in F2 generation in these crops is very difficult at the juvenile stage. DNA markers are reliable, free from environmental influence and can be assayed at any stage of plant growth with any type of tissue. Recent advances in the field of plant molecular genetics have resulted in the development of a series of DNA markers. Restriction fragment length polymorphism (RFLP), (Botstein et al., 1980) random amplified polymorphic DNA (RAPD) (Williams et al., 1990; Welsh and McClelland, 1990), amplified fragment length polymorphism (AFLP), and simple sequence repeats (SSRs), or microsatellites (Weber and May, 1989) are important among them.
