Abstract

Cyanobacteria, although procaryotes, perform oxygenic photosynthesis in a process similar to that observed in the chloroplasts of higher plants but are more amenable to genetic manipulation. The unicellular strains, Anacvstis nidulans R2 and A. nidulans R2-SPc, can be transformed by exogenous plasmid or chromosomal DNA at a high frequency, allowing for molecular genetic manipulations. This laboratory has been concerned with improving the frequency of transformation in A. nidulans and with the construction of hybrid cloning vectors which will allow the controlled expression of genes cloned into these vectors. We report here the construction of three different hybrid cloning vehicles which will allow us to assess the effects of genetic manipulations on the functioning of the photosynthetic apparatus. We also report the use of low iron medium to improve the efficiency of transformation.
