Abstract
On the isolated chloroplast coupling factor CF1, three nucleotide binding sites were found (1) : site 1 binds an ADP molecule which remains bound even after isolation; this ADP is exchanged during incubation of the isolated enzyme with ADP or ATP. Site 2 binds a MgATP molecule; CF1 stored under ammonium sulfate contains no nucleotide on this site. Upon addition of ATP and magnesium ions, this site is filled with MgATP which remains bound even after prolonged incubation with unlabeled nucleotides and/or EDTA. Site 3 binds ADP or ATP in a reversible manner; the dissociation constant is about 2 to 5 μM.
