Identification of a Δ5-Fatty Acid Desaturase from Physcomitrella Patens

Abstract

Very long-chain polyunsaturated fatty acids (VLCPUFA) such as arachidonic (ARA, 20:4ω6) and eicosapentaenoic acid (EPA, 20:5ω3) are considered to be beneficial in the human diet and a sustainable source of VLCPUFA would be highly desirable. The biosynthesis of Δ5-unsaturated VLCPUFA occurs through sequential desaturation and chain-elongation in many eukaryotic organisms, but not in angiosperm plants. A good source to clone the required genes from a plant is the moss Physcomitrella patens with Iipids containing up to 30% ARA and some EPA (Grimsley et al., 1981). Recently we have identified genes encoding a Δ6-desaturase (Girke et al., 1998) and a Δ6-specific elongase from P. patens (Zank et al., in press).
