Abstract

Biodiversity of population of Bacillus thuringiensis, B. licheniformis, and B. megaterium was studied by flow cytofluorometry and other methods. The distributions in size, DNA content and RNA and protein contents of the cells were measured by FACStarPlus flow cytofluotometer (Becton Dickinson). The bacterial neuston contains more cells from the beginning stage of cell division cycle and DNA replication cycle that the cells from the bulk of water. Anabiotic and endotrophic subpopulations of bacilli are accumulated in the water-air interface. The distributions of the cells by DNA content have the different forms depending on the share of C-period within the division cycle. Flow cytofluorometry analysis of the synchronous cultures of bacilli shown that the curves have the patterns with one maximum, two maxima or with one maximum and plateau. The share of cells in B-period and share of exotrophic cells of population are suitable characteristics of the physiological state of populations and can be rapidly determined by the flow cytofluorometry. Their determination may be used for identification of the source and term of biohazard’s release into the environment by the comparance of physiological state in the release point and in the environment; for the determination of the physiological state of biohazard in the natural ecosystems which is important for the risk assessment; and for the determination of the physiological state of biohazard before release into the environment by the comparance of the changes of physiological state in the sampling point(s) of the environment.
