Abstract

During the past few years, several laboratories have reported the successful culture of protoplasts from woody species, a group which has long been considered recalcitrant. A review by McCown and Russell [41] covering the literature through May 1986, listed four woody genera (Citrus, Populus, Santalum, and Ulmus) in which protoplasts had been cultured through whole plant regeneration. Now plants have been regenerated from protoplasts of 46 woody genotypes representing 32 species, 18 genera, and 12 families (Table 1). In many of these cases the protoplast source tissue originated from non-seedling (mature) plants (Table 1). Thus, it will be possible to use protoplast-based genetic engineering to add one or a few desirable traits to already proven varieties so that the development time for a saleable product can be reduced.
