Specificity of Plant-Fungus Interactions: Molecular Aspects of Avirulence Genes

Abstract

A short overview will be presented on genetic and molecular aspects of fungal avirulence genes. The work on Bremia lactucae and Magnaporthe grisea will be discussed. However, emphasis will be on the state of the art concerning avirulence gene Avr9 of the fungal pathogen Cladosporium fulvum, the causal agent of tomato leaf mould, which has been used as a model to study gene-for-gene relationships in our laboratory. A race-specific peptide elicitor, the putative product of Avr9, induces a hypersensitive response (BR) on Cf9 tomato genotypes. Oligonucleotide probes were designed based on the amino acid sequence. With one of these probes a cDNA clone was isolated which encodes the Avr9 elicitor. The mRNA contains an open reading frame of 63 amino acids, with the sequence of the mature elicitor at the C-terminus. Under certain growth conditions the Avr9 mRNA can be induced in liquid shake cultures. The Avr9 gene appears to be a single copy gene which is absent in fungal races which are virulent on tomato Cf9 genotypes.
