Origin and Nature of Somaclonal Variation in Potato

Abstract

A review of data on in vitro cultures of potato shows that a high degree of cytogenetic variation occurs as the result of instability of nuclear processes at segregational, structural and DNA levels. As the differentiated tissues in vivo contain both endoreduplicated and normal cells, (polysomaty), the possibility is suggested that this contributes to in vitro variation. Polyploidy and aneuploidy occur during the early stages of callus induction and persist during callus phase. The data reveal that at these stages, both endoreduplication and endomitosis are important mechanisms of polyploidization, and that chromosome lagging and non-disjunction contribute to the production of aneuploidy. Plants regenerated from protoplasts, cells or tissue cultures showed a wide range of ‘somaclonal variation’ in several commercial varieties, breeding clones and interspecific hybrids. The nature and extent of somaclonal variation is discussed in relation to genotype, expiant source and culture conditions.
