Summary
Two major classes of cytoskeletal proteins characteristic of epithelial cell differentiation, i.e. the constituents of the desmosome and the proteins forming the intermediate-sized filaments (IFs) of the cytokeratin category, occur in the epithelia as well as in the hair-forming cells (trichocytes) of bovine and human hair follicles. Using immunofluorescence microscopy we show the localization of desmoplakin, a representative cytoplasmic component of the desmosomal plaque, and desmoglein, as a representative of a desmosomal transmembrane glycoprotein, in the various epithelial cells and trichocytes of the follicle. Eight major and two minor trichocyte-type cytokeratin polypeptides are described in both species. Double label immunofluorescence microscopy using antibodies specific for trichocytic cytokeratins, in combination with antibodies to certain epithelial cytokeratins, allows the demonstration of a complex pattern of cytokeratin expression in the various cell types contributing to the follicular structure. While certain cells are characterized by the mutually exclusive synthesis of either epithelial or trichocytic cytokeratins, other cells, including basal cells of the matrix in the peripapillary cone and cells of the cuticle, are characterized by specific patterns of co-expression of trichocytic and epithelial cytokeratins, including the non-epidermal cytokeratins 8, 18 and 19 that are normally typical of simple epithelia. The results are summarized in a scheme showing the regional differences of cytokeratin expression in the various elements of the hair follicle.
