Amides, Proteins and Polypeptides

  • Chapter

Abstract

Although there is a good deal of resonance towards a dipolar form, amides do not enolise so that they show a carbonyl absorption, the frequency of which will be influenced to some extent by the dipolar structure with which it can resonate. It will also be affected to a lesser extent by the nature of the R group adjacent to the carbonyl. This absorption is termed the amide I band and is common to all types of amide, including cyclic forms. Its frequency is, however, markedly affected by hydrogen-bonding effects, so that considerable shifts can occur on passing from the solid to solution. This applies with even greater force to the other characteristic amide bands, so that in correlation work on amides and related materials particular attention must be paid to the physical state in which the material is examined.
