Genetic Analysis of the Role of Toxin Production by Pseudomonas syringae pv. syringae in Plant Pathogenesis

Abstract

Pseudomonas syringae pv. syringae strain B301D produces the lipodepsipeptide Phytotoxins, syringomycin and syringopeptin. Analysis of the syringomycin (syr) gene cluster of strain B301D indicated that the toxin is synthesized on multienzymes by a thiotemplate mechanism of peptide synthesis. The syr genes encoding syringomycin synthetases were found to be clustered in an approximately 32-kb region of the chromosome. Specific plant metabolites, as exemplified by certain flavonoid glycosides from cherry leaves, serve as environmental signals sensed by P. s. pv. syringae to activate genes dedicated to syringomycin synthesis. Evidence also is presented that syringomycin and syringopeptin form pores in host membranes and that plant cells are killed as a result of a rapid and sustained flux of cations. The fipopeptide toxins are concluded to be major determinants of bacterial virulence.
