Abstract

Tan necrotic lesions on wheat leaves are one of two possible symptoms of susceptible reactions observed in interactions between isolates of Pyrenophora tritici-repentis and its wheat host. These lesions are produced in an isolate- and wheat host-specific manner. The ability of an isolate to induce tan necrosis (nec+ isolate) on susceptible cultivars was shown to coincide with its ability to produce a host specific toxin in culture. This HST was subsequently purified and shown to be a protein. Using antiserum raised against the toxin we have shown that the toxin is also produced in planta. The antiserum provided the means for isolating the gene encoding the toxin from a cDNA library. Based on the sequence of the toxin gene and the known size of the protein, it is apparent that the toxin is produced as a proprotein and processed to its mature toxin form. A number of post-translational changes may exist but none have been confirmed. The E. coli expressed toxin proprotein has been examined for activity and structure. Using the cloned toxin gene it has been possible to examine the occurrence of the toxin gene or related sequences in a number of isolates of the pathogen and demonstrate that a single copy of the gene appears to be present in races that are nec+ and further that the gene is absent from the genome of necisolates.
