Abstract
With the purpose of evaluating the effect of vinpocetine (Cavinton®), given in a single-dose infusion, on the cerebral metabolism and perfusion of chronic stroke patients, we measured in 12 patients with focal cerebrovascular lesions the regional and global cerebral metabolic rates of glucose with positron emission tomography (PET) before and after a single dose intravenous vinpocetine treatment. The global and regional glucose metabolic rates showed no significant changes before and after Vinpocetine treatment, however, the kinetic constants (k1, k2), related to the facilitated transport of glucose through the blood-brain-barrier in both directions, significantly increased in the whole brain. This increase could be observed in the non-affected hemisphere and in the affected hemisphere outside the infarcted area.
The findings indicate that a single-dose vinpocetine treatment significantly improves the transport of glucose through the blood-brain-barrier in both directions (uptake and release of glucose) in the non-affected hemisphere and in the peri-infarct region of the affected hemisphere.
