Adenovirus Vectors in Functional Genomics

Abstract

Among more than one hundred different serotypes of adenoviruses (Ads) known, 47 can cause usually mild respiratory or gastrointestinal infections in humans (reviewed in Horwitz, 1996). Although they show tropism only for epithelial cells during a regular course of infection, they are able to infect a wide variety of cell types. One of the unique features that has motivated the study of Ads is their ability to replicate at extremely high titers since they can saturate the infected cells with more than 3X104 infectious units (IU)/cell, yielding abundant material for biochemical analysis. After their initial discovery, substantial information has accumulated on their biology and molecular characteristics over the following three decades. This wealth of information has promoted their use as expression vectors initially for recombinant protein production, then as recombinant vaccines, and more recently as gene transfer vectors in animal experiments as well as in human trials (reviewed in Acsadi et al, 1995; Berkner, 1988; Graham and Prevec, 1992; Imler, 1995; Kovesdi et al, 1997; Randrianarison-Jewtoukoff & Perricaudet, 1995; Wiley, Gene Therapy web site).
