Genetic Enhancement of Protein Productivity of Animal Cells by Oncogenes

Abstract

Effects of oncogenes such as c-myc,c-fos, v-myb, and c-Ha-ras were examined on the exogenous protein production by cultured animal cells. Several plasmids expressing oncogenes were constructed as effector plasmids and a plasmid containing the IL-6 gene regulated by the SRa promoter as a reporter plasmid. In transient transfection experiments, only c-myc oncogene stimulated the IL-6 production several times in BHK21 cells. To examine the effects of oncogenes on the permanent expression of the IL-6 gene, the IL-6 gene was introduced into BHK-21 cells. Highly productive clones that secreted 10 times more IL-6 than parental clones were obtained by further transfection of the recombinant BHK-21 cells with oncogenes, suggesting that oncogene products activated the SRa promoter, resulting in the high production of IL-6. These results suggests that it will be possible without gene amplification to get highly productive clones in a short time by using oncogenes for activation of the promoter that regulates a product gene.
