Electrically controlled culture of MKN45 cells in serum-free medium

Abstract

Specific protein production of mammalian cells have been promoted by applying a low constant potential to the surface of an electrode on which cells were cultured in serum-free medium. A human carcinoma cell line, MKN45 cells, were cultured on a platinum-coated plate electrode and carbon-mesh electrode with an applied potential below 0.6 V vs. Ag/AgCl in serum-free medium. The carcinoembryonic antigen (CEA) production was dependent on the electrode potential and was rather enhanced at 0.4 V vs. Ag/AgCl on both types of electrode. This optimum potential of 0.4 V vs. Ag/AgC1 agreed with the potential that maximally promoted the CEA production in fetal bovine serum (FBS) containing medium. This phenomenon could led us to design a new type of bioreactor which would control the protein production by electrical effects.
