Continuous Culture with Cell Precipitation for Recombinant Protein Production

Abstract

A continuous bioreactor using a spinner flask equipped with cell precipitator was applied to cultivate insect cells. Maximum viable density of insect cells in this bioreactor system reached 1.38 x 106 cells/ml at the dilution rate of 0.005 hr-1. Recombinant protein production by cells infected with a genetically-modified baculovirus was also demonstrated. The maximum β-galactosidase synthesis of 7500 units per reactor volume was achieved at the dilution rate of 0.011 hr-1.
