Testing Primary Charge Separation in Photosynthetic Reaction Centers with External Electric Fields

Abstract

The mechanism of primary charge separation in photosynthetic reaction centers is yet unclear. The results and shortcomings of directly tracing this process spectroscopically are outlined. Then the expected influence of an electric field on this process is discussed. The method of determining the orientation of the dipole moment of the primary radical pair from the Dichroic Excitation spectrum of eLectric Field modulated Yield of the prompt fluorescence (DELFY) will be introduced promising direct access to the identity of the primary electron acceptor. The results of low temperature steady state DELFY experiments on RCs from Rb.sphaeroides R-26 are presented pointing to single step formation of P+H-. The implications of time resolved fluorescence and absorption measurements in an electric field on this result are demonstrated and discussed in the frame of various models.
