Monoclonal Antibodies as Tools in Early Detection of Plant Pathogens

Abstract

Immunoenzymatic tests are currently finding broad use in phytopathology. A hybridoma line secreting monoclonal antibody against Erwinia amylovora, the agent of fireblight, was developed by fusion of spleen cells of an immunized BALB/c mouse with P3x63-Ag 8.653 mouse myeloma cells. The monoclonal antibody was found to be specific for E. amylovora revealing no cross reaction with other Erwinia strains. The antibody was of IgG2A sub isotype. Its antigen was found to be a poly-peptide of 20000 Mr. The enzyme-linked immunoadsorbent assay (ELISA) system developed by use of the monoclonal antibody permitted antigen detection with a sensitivity limit of 103–104 bacteria.
